Challenges and Pitfalls in Clinical Translation of 2D Graphene Materials as Nanoplatforms for Canonical and Non-Canonical Therapeutics
The use of nanomaterials in medicine has been growing at an unprecedented rate for a variety of therapeutic, diagnostic or combinatory applications. The clinical translation of advanced materials that are being discovered or synthetically engineered is considered by many the critical factor to determine the ´success´ or ´failure´ in nanomedicine. However, the road to clinical translation of advanced materials, even at the very early development stages, is full of challenges and pitfalls that commonly preclude their translation to advanced stages such as Phase II and above clinical trials that assess efficacy. In this presentation, our decade-long efforts in the clinical translation of graphene oxide two-dimensional (2D) nanosheets as nanoplatforms for the delivery of ‘canonical’ (i.e. molecules with biological activity) and ‘non-canonical’ (i.e. electrical current in the form of neuromodulation) will be discussed. Recent progress in the development of clinically trialed graphene oxide nanosheets will be described, with focus on two very different first-in-human (FIH) clinical investigation studies undertaken recently using graphene-based technologies.
