Engineering Stimuli-Responsive Nanocarriers for Cancer Diagnosis and Immunotherapy
Because hypoxia is a common feature of solid tumors and has been strongly associated with resistance against therapies, developing hypoxia-sensitive nanocarriers would improve the therapeutic outcomes. As the pathological features of hypoxia in esophageal squamous cell carcinoma (ESCC) tumors remain unclear, we hypothesized that identifying hypoxia in ESCC tumors would cooperatively afford benefits for developing hypoxia-responsive nanocarriers for efficient drug delivery and targeted therapy. Herein, we identified high T-lymphokine-activated killer cell-originated protein kinase (TOPK) expression in human ESCC tumors and its close correlation with patient prognosis and clarified hypoxia as another pathological hallmark of ESCC tumors. We developed hypoxia-sensitive nanoparticles encapsulating TOPK inhibitor OTS964 and photosensitizer chlorin e6 for imaging-directed precision therapy of ESCC tumors.
