Title: Lyophilized lymph nodes for improved delivery of chimeric antigen receptor T cells
Many challenges impede chimeric antigen receptor (CAR) T cell activity against solid tumours. Here we described a strategy of delivering CAR T cells via patient-derived lyophilized lymph nodes (L-LNs) to inhibit postoperative tumour recurrence. LNs provide a specialized structure and milieu that supports T and B lymphocyte homeostasis. Essential nutrients and growth factors enhance cell recruitment and survival. Regional LNs are commonly removed during dissection of the primary tumour to provide prognostic information and identify nodal metastases. Thus, LNs could be used to generate a biocompatible and autologous material to deliver CAR T cells. We found that L-LNs create a favorable environment for T cell functions, maintaining their viability, promoting proliferation, and sustaining anti-tumour effects. Compared to traditional hyaluronic acid-based hydrogels, L-LNs are more effective in delivering CAR T cells and therefore preventing tumor recurrence.
